Characteristics | This reagent is intended to be used in a labeled streptavidin-biotin immunoenzymatic antigen detection system. This technique involves the sequential incubation of the specimen with an unconjugated mouse primary antibody specific to the target antigen, a biotinylated secondary antibody which reacts with the primary antibody, enzyme- labeled streptavidin, and substrate-chromogen. |
Protocol |
1. Deparaffinize and rehydrate formalin-fixed paraffin-embedded tissue section. 2. Perform appropriate pretreatment and blocking if required. Wash 3 times in buffer. 3. Apply primary antibody and incubate according to manufacturer’s protocol. 4. Wash 4 times in buffer. Apply Biotinylated Goat Anti-Mouse and incubate 10-15 minutes at room temperature. Wash 4 times in buffer. 5. Apply appropriate enzyme-labeled streptavidin and incubate according to the manufacturer’s instructions. 6. Rinse 4 times in buffer. Apply chromogen and substrate according to manufacturer’s instructions. 7. Apply counterstain according to manufacturer’s instructions. 8. Dehydrate and coverslip or use appropriate mounting medium. |

Storage Comment | The user must validate any other storage conditions. The reagents must be returned to the storage conditions identified above immediately after use. When properly stored, the reagent is stable to the date indicated on the label. Do not use the reagent beyond the expiration date. There are no definitive signs to indicate instability of this product, therefore, positive and negative controls should be tested simultaneously with unknown specimens. |
