Specificity Protease Inhibitor Cocktail is a broad-spectrum cocktail designed to prevent proteolytic degradation during lysis and extraction procedures from mammalian cells or tissues.
Characteristics The cocktail contains a broad-spectrum of inhibitors targeting cysteine proteases, serine proteases, aspartic acid proteases, and aminopeptidases. In addition to being ready-to-use, the Protease Inhibitor cocktail is also EDTA-free. The cocktail is supplied as a 100X stock solution in a liquid format that has been stabilized in dimethylsulfoxide (DMSO) for improved accuracy, solubility, and ease of use in comparison to traditional tablets.
- AEBSF, 104 mM - Serine Proteases
- Aprotinin, 80 μM - Serine Proteases
- Bestatin, 4 mM - Aminopeptidases
- E-64, 1.4 mM - Cyseine Proteases
- Leupeptine, 2 mM - Serine and Cysteine Proteases
- Pepstatin A, 1.5 mM - Asparic Acid Proteases
Comment

The cocktail solution inhibits aminopeptidases and cysteine, serine and aspartic acid proteases. Protease Inhbitor Cocktail is provided EDTA-free, as a ready-to-use 100X stock solution in DMSO. The cocktail composition and inhibitor targets are listed below:


  • AEBSF, 104 mM - serine proteases
  • Aprotinin, 80 μM - serine protease
  • Bestatin, 4 mM - aminopeptidases
  • E-64, 1.4 mM - cysteine proteases
  • Leupeptin, 2 mM - serine and cysteine proteases
  • Pepstatin A, 1.5 mM - aspartic acid proteases

It is recommended to use 10 μl of the cocktail solution for every 1 ml of cell lysate. Since protease levels may vary among cell and tissue types, it may be necessary to increase the concentration of inhibitors.
